The competitive inhibitors of cyclooxygenase (COX), the enzyme that facilitates the bioconversion of arachidonic acid to inflammatory prostaglandins (PGs), are non-steroidal anti-inflammatory medicines (NSAIDs). The usage of them is linked to side effects like renal and gastrointestinal toxicity. While the undesirable side effects of NSAIDs result from the suppression of COX-1 activity, the therapeutic anti-inflammatory impact is provided by the inhibition of COX-2. Therefore, it was assumed that side effects would be lower with more specific COX-2 inhibitors. As safer NSAIDs with an enhanced stomach safety profile, a variety of selective COX-2 inhibitors (rofecoxib, celecoxib, valdecoxib, etc.) were developed.
